A silver Mercedes travelling towards Kennford left the carriageway, collided with a tree in the grassed central reservation and ended up on its side.
An off-duty Exeter detective who was travelling behind the vehicle stopped at the scene and administered first aid on the male driver, who was the only person in the car.
She was assisted by a member of the public and the first officer on the scene before paramedics arrived.
The man, believed to be in his 60s and from the Totnes area, was taken to Royal Devon and Exeter Hospital where he later died. Formal identification has not yet taken place.
The road was closed for six hours while police investigations were carried out.
Officers from Exeter Serious Collisions Investigation Unit have launched an investigation into the collision and are appealing for witnesses to contact them.
Anyone with information is asked to contact the collisions unit by telephoning 101, quoting log number 562 of 14/01/13.
